DOMS is not only when you're beginning or after a break, but as a result of fatiguing the muscle. It doesn't mean you'll have crippling DOMS, but you'll be stiff and definitely know you've hit a muscle group hard.
The human body is great at adapting. If you do something everyday, it will get stronger. Making it harder to fatigue. In a way bodybuilding is the opposite of this. You don't want your body to adapt; you want it to be hit hard, so it rebuilds larger.
